Overview

CD33.CD28z CAR T cells are an investigational autologous T-cell therapy engineered to express a chimeric antigen receptor (CAR) targeting CD33 (Siglec-3), a transmembrane receptor highly expressed on the surface of acute myeloid leukemia (AML) blasts and leukemic stem cells. The CAR construct consists of a CD33-specific single-chain variable fragment (scFv) linked to a CD28 costimulatory domain and a CD3-zeta signaling endodomain. This second-generation CAR design aims to enhance T-cell activation, proliferation, and persistence upon antigen engagement. Developed by the Center for Cell and Gene Therapy at Baylor College of Medicine, these CAR-T cells are being evaluated in clinical trials for patients with relapsed or refractory AML, both as a standalone therapy and as part of combinatorial strategies to mitigate antigen escape and address disease heterogeneity.
