CD33KO-Loop CAR-iNK cells are an experimental allogeneic cell therapy consisting of natural killer (NK) cells derived from human induced pluripotent stem cells (iPSCs). These cells are engineered with a bivalent 'Loop CAR' that simultaneously targets CD33 and mesothelin (MSLN), antigens frequently expressed in acute myeloid leukemia (AML). To prevent fratricide (self-killing) caused by the expression of CD33 on the NK cells themselves, the CD33 gene is knocked out using CRISPR/Cas12i technology. This modification enhances the expansion and persistence of the therapeutic cells. The therapy is designed to overcome tumor antigen escape and reduce the toxicities associated with traditional autologous CAR-T therapies, providing a standardized, off-the-shelf immunotherapy product.
