Overview

CD34+ purged stem cells refer to an autologous hematopoietic stem cell graft that has undergone ex vivo processing to enrich for CD34+ progenitor cells while depleting contaminating malignant cells. This process, often performed using immunomagnetic separation devices like the CliniMACS system, is utilized in autologous stem cell transplantation (ASCT) for hematologic malignancies such as peripheral T-cell lymphoma (PTCL). Since most PTCL cells do not express the CD34 antigen, positive selection of CD34+ cells effectively "purges" the graft of tumor cells, potentially reducing the risk of relapse post-transplant. Clinical data suggests that using purged grafts can improve overall survival and disease-free survival in high-risk PTCL patients compared to unpurged grafts, without significantly delaying hematopoietic engraftment.
