CD34+ selected donor cells are a purified population of hematopoietic stem and progenitor cells (HSPCs) derived from an allogeneic donor's peripheral blood or bone marrow. The production process typically utilizes immunomagnetic separation technology, such as the CliniMACS system, to selectively enrich for cells expressing the CD34 surface antigen while significantly depleting T-lymphocytes and other mature immune cells. This therapy is primarily indicated for patients undergoing allogeneic hematopoietic stem cell transplantation (HSCT) for hematologic malignancies, such as acute myeloid leukemia (AML). By drastically reducing the T-cell content of the graft, the therapy is designed to minimize the incidence and severity of Graft-versus-Host Disease (GvHD), a potentially fatal immune complication where donor T-cells attack recipient tissues, without requiring intensive pharmacological immunosuppression. The purified CD34+ cells retain the capacity to home to the bone marrow and differentiate into all lineages of the hematopoietic and immune systems, facilitating full engraftment and immune reconstitution.
