This investigational cell therapy approach, developed by the National Heart, Lung, and Blood Institute (NHLBI), involves the use of allogeneic peripheral blood stem cells (PBSC) obtained from partially matched related donors. The therapeutic platform utilizes positive selection of CD34+ hematopoietic progenitor cells combined with extensive T-cell depletion of the graft. This dual mechanism is designed to facilitate hematopoietic reconstitution while significantly reducing the risk of severe graft-versus-host disease (GVHD), which is a primary complication in mismatched allogeneic transplantation. The therapy has been evaluated in Phase II pilot studies as part of a transplantation platform for patients with high-risk hematologic malignancies, including acute myeloid leukemia (AML), acute lymphocytic leukemia (ALL), chronic myelogenous leukemia (CML), myelodysplastic syndromes (MDS), and myeloproliferative disorders.
