CD34-selected autologous peripheral blood stem cells is an investigational cell therapy developed by the Fred Hutchinson Cancer Center for the treatment of chronic lymphocytic leukemia (CLL). The procedure involves the collection of a patient's own peripheral blood stem cells (PBSCs) followed by an ex vivo selection process to enrich for CD34+ hematopoietic stem and progenitor cells. This selection serves to purge the graft of contaminating malignant CLL cells (B-lymphocytes), which are typically CD34-negative, thereby reducing the risk of re-infusing cancer cells. Following myeloablative conditioning with high-dose chemotherapy and total body irradiation, the purified stem cells are transplanted back into the patient to restore bone marrow function and hematopoiesis.
