Overview

CD34-selected progenitor cells refer to a cellular therapy product consisting of purified CD34+ hematopoietic stem and progenitor cells (PBPCs) derived from donor peripheral blood. Developed and studied at M.D. Anderson Cancer Center, this approach is utilized in allogeneic stem cell transplantation for patients with advanced hematological malignancies, including leukemia, lymphoma, and myelodysplastic syndrome. The manufacturing process involves ex vivo T-cell depletion through positive CD34+ selection, typically using the CliniMACS device, to deliver a 'megadose' of stem cells. This technique aims to facilitate donor-derived neutrophil and platelet engraftment while significantly reducing the incidence of acute and chronic graft-versus-host disease (GvHD) by removing the T-cells responsible for the immune attack on the recipient's tissues.
