CD34 Stem Cell Selection Therapy is an investigational cell-based biological therapy developed by Dr. Diane George at Columbia University Medical Center. The therapy involves the ex vivo processing of allogeneic peripheral blood stem cells from matched or partially matched family or unrelated adult donors using the CliniMACS PLUS system (Miltenyi Biotec). By selectively enriching for CD34+ hematopoietic stem cells and depleting T-cells (CD3+ cells), the procedure aims to facilitate successful engraftment and immune reconstitution in patients with non-malignant disorders while significantly reducing the risk of graft-versus-host disease (GVHD). It is primarily being evaluated in children, adolescents, and young adults with a wide range of conditions, including bone marrow failure syndromes, primary and acquired immunodeficiencies, histiocytic disorders, and hemoglobinopathies such as sickle cell disease and beta-thalassemia.
