CD37 CAR-T is an adoptive cell therapy consisting of T cells genetically engineered to express a chimeric antigen receptor (CAR) targeting the CD37 antigen. CD37 is a tetraspanin protein predominantly expressed on mature B cells and is also found on certain myeloid cells, making it a therapeutic target for B-cell malignancies (such as non-Hodgkin lymphoma and chronic lymphocytic leukemia) and acute myeloid leukemia (AML). The CAR construct typically includes an extracellular single-chain variable fragment (scFv) derived from an anti-CD37 antibody, a transmembrane domain, and intracellular costimulatory domains (e.g., 4-1BB or CD28) plus a CD3-zeta signaling domain. Upon recognition of CD37-positive tumor cells, these engineered T cells proliferate and mediate targeted cell lysis through the release of perforins, granzymes, and pro-inflammatory cytokines. In preclinical studies, CD37 CAR-T has been used both as a standalone therapeutic candidate and as a component of bispecific CAR constructs to overcome antigen heterogeneity.
