CD37-Siglec6 bispecific CAR-T is a preclinical-stage chimeric antigen receptor T-cell (CAR-T) therapy designed to simultaneously target CD37 and Sialic acid-binding Ig-like lectin 6 (Siglec-6), two antigens highly expressed on leukemia stem cell (LSC) subpopulations in acute myeloid leukemia (AML). Developed as an academic research project at Hangzhou Medical College, this therapy aims to overcome challenges in relapsed/refractory pediatric AML, such as antigen heterogeneity and off-tumor toxicity. The CAR constructs are delivered via lentiviral vectors. Preclinical studies in NSG mouse xenograft models have demonstrated that the bispecific approach enhances anti-leukemic activity, improves T-cell persistence, and reduces exhaustion compared to single-target CAR-T cells, while maintaining a favorable safety profile regarding normal hematopoiesis.
