CD37.GS4L CAR-T is an experimental third-generation chimeric antigen receptor (CAR) T-cell therapy targeting the CD37 antigen, which is expressed in various B-cell and T-cell malignancies. The construct features a specific 20-amino acid glycine-serine linker (GS4L) within the single-chain variable fragment (scFv), which has been shown to provide superior flexibility and binding affinity compared to other linker types. The intracellular signaling domain incorporates dual costimulatory molecules, CD28 and CD40, alongside the CD3ζ chain to enhance T-cell activation and persistence. Developed by researchers at Prince of Songkla University and Nagoya University, this CAR-T has demonstrated potent anti-tumor activity in preclinical models of Burkitt's lymphoma, mantle cell lymphoma, multiple myeloma, and cutaneous T-cell lymphoma, while showing reduced fratricide and improved central memory T-cell preservation.
