Overview

CD371-CAR T cells (specifically the CD371-SAVVYz-IL18 construct) is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy designed for the treatment of relapsed or refractory acute myeloid leukemia (AML). The therapy targets CD371 (also known as CLL-1 or CLEC12A), a surface antigen highly expressed on AML blasts and leukemic stem cells but absent on healthy hematopoietic stem cells. The construct features a fully human scFv and a mutated CD28 costimulatory domain, termed 'SAVVY', which is engineered to minimize T-cell exhaustion and improve persistence. Additionally, the cells are modified to constitutively secrete interleukin-18 (IL-18), a potent immunomodulatory cytokine that enhances CAR T-cell cytotoxicity and stimulates host immune responses within the tumor microenvironment. It is currently being evaluated in Phase 1 clinical trials.
