**CD38-CAR-T** is an investigational autologous T cell therapy in which a patient’s own T cells are genetically engineered to express a chimeric antigen receptor (CAR) targeting CD38, a transmembrane glycoprotein highly expressed on the surface of malignant plasma cells in multiple myeloma (MM) and in acute myeloid leukemia (AML). The engineered CAR enables T cells to recognize and specifically eliminate CD38-positive tumor cells by binding to CD38, followed by T cell activation and targeted cytotoxicity. CAR-T-38 cells exhibit efficient antitumor activity in vitro and in vivo, inducing apoptosis of tumor cells through cytokine secretion (e.g., IFN-γ, TNF-α), and proliferate robustly upon CD38-positive target engagement. The technology draws on antibody-derived single-chain variable fragments fused to T cell activation domains and utilizes retroviral transduction for gene delivery. CD38-CAR-T is under clinical investigation for relapsed and refractory MM and AML, with some case series showing potential complete remissions and manageable side effects.
