CD38-IN-78c is a potent, selective, reversible, and uncompetitive small molecule inhibitor of the glycoprotein enzyme CD38, belonging to the thiazoloquin(az)olin(on)e class. CD38-IN-78c inhibits CD38-mediated hydrolysis of NAD+ and related metabolites, leading to elevated tissue NAD+ levels, improved metabolic function, and protection against tissue damage in animal models. It has shown beneficial effects in models of ischemia/reperfusion injury, age-related metabolic decline, and autoimmune conditions such as rheumatoid arthritis and collagen-induced arthritis. Mechanistically, 78c is reported to ameliorate mitochondrial dysfunction, modulate immune cell profiles, and improve exercise capacity and frailty parameters in aged mice. The compound is used mainly in preclinical research, and its development is most closely associated with Dr. Eduardo N. Chini's team at the Mayo Clinic.
