Drug intelligence / Profile preview
CD38-LMP1 tandem CAR-T cells are second-generation chimeric antigen receptor (CAR) T cells engineered to simultaneously target CD38 and latent membrane protein 1 (LMP1). Developed by researchers at the First Affiliated Hospital of Zhengzhou University and Baylor College of Medicine, these tandem CAR-T cells are designed to treat natural killer/T-cell lymphoma (NKTCL), an aggressive hematologic malignancy strongly associated with Epstein-Barr virus (EBV) infection. By targeting both CD38 (highly expressed on NKTCL cells) and LMP1 (an EBV-encoded oncogenic protein), the tandem CAR-T cells aim to enhance cytolytic activity and prevent antigen escape, a common mechanism of resistance in single-target CAR-T therapies. Preclinical studies in vitro and in vivo have demonstrated robust anti-tumor activity, target-dependent activation, and inflammatory cytokine production.
