CD38-SADA is a bispecific recombinant fusion protein designed as a pre-targeted radioimmunotherapy (PRIT) agent for the treatment of relapsed or refractory non-Hodgkin lymphoma (NHL). It utilizes Y-mAbs Therapeutics' proprietary Self-Assembling and Disassembling Antibody (SADA) technology platform, which enables a two-step therapeutic approach. In the first step, non-radiolabeled tetrameric forms of the protein are administered and bind with high avidity to the tumor-expressed antigen, lymphocyte differentiation antigen CD38. Unbound proteins disassemble into monomers that are rapidly cleared by renal excretion. In the second step, a radioactive payload—typically Lutetium 177-DOTA—is infused and binds specifically to tumor-localized antibody constructs for targeted irradiation of cancer cells while minimizing off-target toxicity. The construct was developed using technology licensed from Memorial Sloan Kettering Cancer Center and MIT[1][2][4][5][6][7][9].
