CD38xNKp30 is a novel bispecific natural killer cell engager (NKCE) designed for the treatment of multiple myeloma. Developed by researchers at the University Medical Center Schleswig-Holstein, the molecule is an IgG-like construct that combines a Fab fragment derived from the anti-CD38 antibody daratumumab with a camelid-derived nanobody targeting the activating receptor NKp30 on natural killer (NK) cells. By simultaneously binding CD38 on malignant plasma cells and NKp30 on NK cells, the drug facilitates the recruitment and activation of NK cells to the tumor site, inducing potent tumor cell killing. The construct can be engineered with either a silent or an active Fc domain; the active Fc domain further enhances anti-myeloma activity by co-engaging Fcγ receptors to induce antibody-dependent cellular phagocytosis (ADCP).
