Drug intelligence / Profile preview
CD3:B7H3 bispecific T cell engager is a class of bispecific antibodies designed to redirect T cells to eliminate B7-H3 (CD276)-expressing tumor cells. These molecules typically feature two binding domains: one targeting CD3, a subunit of the T-cell receptor complex on T lymphocytes, and another targeting B7-H3, an immune checkpoint protein overexpressed in various solid tumors, including glioblastoma, medulloblastoma, and sarcomas. By simultaneously binding to both targets, the engager physically bridges T cells and tumor cells, triggering T-cell activation, proliferation, and directed cytotoxic activity against the cancer cells. Multiple candidates utilizing this mechanism, such as MacroGenics' obrindatamab (MGD009) and Takeda's TAK-280, have been investigated in preclinical and clinical settings.
