Overview

CD3x15B6 is a bispecific T-cell engager (BiTE) designed to target mesothelin (MSLN) expressing solid tumors. Unlike traditional MSLN-targeted therapies that bind to the distal N-terminal domain (which is frequently shed by proteases in the tumor microenvironment), CD3x15B6 targets the 15B6 epitope located in the membrane-proximal, protease-sensitive region that remains cell-bound. By simultaneously binding to CD3 on T cells and the non-shed region of MSLN on cancer cells, the antibody facilitates T-cell activation and directed cytotoxicity. Preclinical studies have demonstrated that CD3x15B6 maintains its cytotoxic activity in the presence of shed MSLN and promotes significant anti-tumor activity and immune cell activation in models of mesothelioma, colon, and breast cancer.
