Overview

CD3xBCMA-bsAb is a preclinical-stage bispecific antibody (bsAb) developed by Protheragen for the treatment of multiple myeloma. As a T-cell engager, the molecule is designed to simultaneously bind to the B-cell maturation antigen (BCMA), which is highly and selectively expressed on malignant plasma cells, and the CD3 epsilon subunit of the T-cell receptor complex on cytotoxic T cells. By cross-linking these two targets, the antibody facilitates the formation of an immunological synapse, leading to T-cell activation and the subsequent directed lysis of BCMA-expressing myeloma cells via perforin- and granzyme-mediated pathways. This therapeutic strategy aims to harness the patient's own immune system to eliminate cancer cells, addressing the unmet need in relapsed or refractory multiple myeloma.
