CD3xCD28 ΔCD38

CD3xCD28 ΔCD38 is a trispecific T-cell engager (TCE) isotype control designed with a null or inactivated CD38-binding arm while retaining functional binding domains for CD3 and CD28. Developed by Sanofi as a research tool to evaluate the contribution of CD28 co-stimulation in the context of trispecific antibodies like SAR442257, it helps differentiate between tumor-killing activity driven by CD38 targeting versus that mediated by CD28 expression on target cells (such as multiple myeloma cells). In preclinical studies, it has been used to demonstrate that CD28-mediated co-stimulation can independently drive T-cell activation and tumor cell lysis when the target cells express high levels of CD28.
