CD3xSS1 is an experimental bispecific T-cell engager (TCE) antibody developed by researchers at the National Cancer Institute (NCI). It is designed to target mesothelin (MSLN)-expressing solid tumors, including mesothelioma, pancreatic, and ovarian cancers. The antibody functions by simultaneously binding to the CD3 receptor on T cells and the SS1 epitope of mesothelin on tumor cells, thereby bridging the two cell types and inducing T-cell activation and directed cytotoxicity. The SS1 epitope is located on the distal N-terminal domain of mesothelin, a region that is susceptible to proteolytic cleavage and shedding into the tumor microenvironment. Consequently, high concentrations of shed MSLN can act as a decoy, binding to CD3xSS1 and inhibiting its anti-tumor activity, a limitation that has led to the development of newer constructs targeting non-shed regions of the protein.
