CD3xTRP1 bispecific antibody is a preclinical-stage T-cell engager designed to treat TRP1-positive malignancies, such as melanoma. It simultaneously binds to the CD3 epsilon subunit on T cells and Tyrosinase-Related Protein 1 (TRP1/TYRP-1) on tumor cells. This dual binding facilitates the formation of an immunological synapse, leading to T-cell activation and the release of cytotoxic granules (perforin and granzymes) to induce tumor cell lysis independently of the T-cell receptor's native specificity. Developed through collaboration between Radboud University Medical Center and Leiden University Medical Center, and studied in combination with decoy-resistant IL-18 (ST-067) by Simcha Therapeutics, the antibody is typically engineered with an inert Fc region to prevent non-specific activation via Fc receptors.
