CD4-ADC is a preclinical-stage antibody-drug conjugate (ADC) platform being developed by TaiMed Biologics for HIV treatment/cure and autoimmune diseases. The platform utilizes TaiMed's second-generation CD4-targeting monoclonal antibody, TMB-365 (an engineered, long-acting variant of ibalizumab), as the targeting vehicle to deliver therapeutic payloads directly into CD4-positive immune cells. For HIV cure research, the antibody is conjugated to small-molecule antiretroviral payloads, such as the integrase inhibitor derivative INI-HIV-015 (forming the candidate TMB-365-015), or latency reversal agents (LRAs) and targeted cell-kill activators (TACK) to eliminate latent HIV reservoirs. For autoimmune diseases (such as rheumatoid arthritis, multiple sclerosis, Crohn's disease, psoriasis, and systemic lupus erythematosus), the platform (referred to as TMB-365-IMMUNE ADC) delivers immunomodulatory payloads (e.g., JAK inhibitors) directly to overactivated CD4+ T cells, minimizing systemic immunosuppression and toxicity.
