Overview

CD4-IgG2 (also known as PRO 542) is a recombinant fusion protein designed to neutralize HIV-1 by preventing the virus from attaching to the host cell. It consists of four copies of the D1D2 domains of human CD4 fused to an IgG2 Fc region, creating a tetravalent molecule with high avidity for HIV-1 gp120. This structure allows it to potently neutralize primary HIV-1 isolates by binding and cross-linking viral envelope proteins, thereby blocking viral entry into monocytes/macrophages and T cells. The drug has demonstrated antiviral activity in both adult and pediatric patients with HIV-1 infection in early-phase clinical trials. Its mechanism is similar to that of anti-CD4 binding site antibodies and may also induce gp120 shedding from virions[1][3][5][6].
