CD4-targeted CD19 CAR fusosome is an investigational **in vivo CAR-T gene therapy** platform from Sana Biotechnology designed to generate anti-CD19 CAR-T cells directly inside the patient rather than through ex vivo cell manufacturing. The product consists of a **paramyxovirus-based integrating fusosome vector** engineered to selectively bind **CD4-positive T cells** using anti-CD4 scFv or VHH targeting domains and deliver a transgene encoding a **CD19-directed chimeric antigen receptor** containing **4-1BB** and **CD3 zeta** signaling domains. After in vivo transduction of resting and activated CD4-positive T cells, the resulting engineered T cells are intended to recognize and eliminate **CD19-positive B cells** and B-cell malignancies, with proposed applications including **B-cell acute lymphoblastic leukemia** and other CD19-positive cancers.
