Overview

CD40.HIVRI.Env is a first-in-class prophylactic HIV vaccine candidate designed to target dendritic cells by combining a humanized anti-CD40 monoclonal antibody fused with HIV envelope (Env) antigens. The vaccine specifically targets the CD40 molecule expressed on the surface of dendritic cells, enabling direct delivery of HIV Env antigens to these key antigen-presenting cells. This approach aims to stimulate both humoral and cellular immune responses against HIV by harnessing the intrinsic immunostimulatory properties of dendritic cell targeting. In Phase I clinical trials, CD40.HIVRI.Env demonstrated safety and induced early, strong, and sustained immune responses in healthy volunteers. The vaccine is adjuvanted with Poly-ICLC (Hiltonol) and has been evaluated alone or in combination with DNA-HIV-PT123 in prime-boost strategies[1][2][3][4][5][6][7][8].
