CD40.HVac is a first-in-class immunotherapy designed to target dendritic cells by combining a humanized anti-CD40 monoclonal antibody with the E6/E7 oncoproteins from human papillomavirus type 16 (HPV16). This fusion protein acts as a therapeutic vaccine, aiming to induce robust and specific immune responses against HPV16-associated cancers. The mechanism leverages the activation of dendritic cells via CD40 engagement, enhancing antigen presentation and stimulating both CD4+ and CD8+ T cell responses against tumor antigens. The drug is being developed primarily for HPV16-positive oropharyngeal carcinoma but has potential applications in other HPV-induced cancers, prostate cancer, infectious diseases such as COVID-19 (prophylactic vaccine), and HIV (therapeutic vaccine). It is currently in Phase I/II clinical trials for HPV16+ oropharyngeal cancer[6][7][8][9].
