CD40.Pan.CoV vaccine adjuvanted with Hiltonol is an experimental pan-coronavirus vaccine candidate developed by ANRS, Emerging Infectious Diseases and the Vaccine Research Institute (VRI). The vaccine component, CD40.Pan.CoV, is a protein subunit vaccine consisting of a monoclonal antibody fragment targeting the CD40 receptor on antigen-presenting cells (APCs) fused to conserved coronavirus antigens. This targeting mechanism is designed to enhance the activation of dendritic cells and subsequent T-cell and B-cell immune responses. The vaccine is formulated with Hiltonol (Poly-ICLC), a synthetic double-stranded RNA that acts as a Toll-like receptor 3 (TLR3) and MDA5 agonist, providing a potent adjuvant effect to stimulate innate immunity. It is currently being investigated in the Phase 1 SOLVE-01 clinical trial as a booster dose in healthy adults previously vaccinated with authorized mRNA COVID-19 vaccines.
