CD40.S (also known as VRI-02) is a next-generation protein subunit vaccine candidate developed by the Vaccine Research Institute (VRI) and ANRS Maladies Infectieuses Emergentes. The vaccine utilizes a unique targeting strategy consisting of a humanized monoclonal antibody directed against the CD40 receptor, which is covalently fused to the SARS-CoV-2 Spike protein. CD40 is a critical receptor expressed on professional antigen-presenting cells, particularly dendritic cells. By binding to CD40, the vaccine not only delivers the viral antigen directly to the cells responsible for initiating the immune response but also acts as an adjuvant by triggering dendritic cell maturation and activation. This mechanism is intended to enhance the magnitude, quality, and durability of T-cell and B-cell memory responses, making it a promising candidate for use as a booster to protect against emerging SARS-CoV-2 variants.
