CD40HVac is a therapeutic vaccine designed for the treatment of HPV16-associated malignancies, specifically oropharyngeal squamous cell carcinoma. It consists of a humanized IgG4 monoclonal antibody targeting the CD40 receptor on dendritic cells, which is fused to the HPV16 E6 and E7 oncoproteins. This design allows for the direct delivery of viral antigens to dendritic cells, promoting a robust T-cell-mediated immune response. The vaccine is administered in combination with **poly-ICLC** (Hiltonol), a synthetic double-stranded RNA that acts as an immune adjuvant by stimulating Toll-like receptor 3 (TLR3) and MDA5 pathways. Developed by LinKinVax (formerly EnnoDC) in collaboration with Gustave Roussy, the combination is currently being evaluated in Phase I/IIa clinical trials for patients in remission following curative treatment for HPV16-positive oropharyngeal cancer.
