Overview

The CD40L bystander vaccine is a cell-based cancer immunotherapy designed to stimulate the immune system against tumors. It typically consists of irradiated tumor cells combined with "bystander" cells (such as K562) that have been genetically engineered to express both granulocyte-macrophage colony-stimulating factor (GM-CSF) and CD40 ligand (CD40L). The GM-CSF recruits and activates dendritic cells at the vaccination site, while CD40L enhances antigen presentation and T-cell activation. This dual action aims to induce a robust anti-tumor immune response mediated primarily through dendritic cell activation and subsequent T-cell priming. The approach has been investigated in clinical trials for metastatic lung adenocarcinoma and other cancers[1][2][3].
