A CD44 neutralizing antibody is a monoclonal antibody designed to bind to the CD44 cell surface protein, a glycoprotein that serves as a primary receptor for hyaluronic acid and other ligands such as osteopontin. CD44 is frequently overexpressed in various malignancies, including glioblastoma multiforme (GBM), and is recognized as a marker for cancer stem-like cells (CSCs). By binding to the extracellular domain of CD44 (or specific isoforms like CD44v6), these antibodies block ligand-receptor interactions and inhibit downstream oncogenic signaling pathways, such as the PI3K/AKT and MAPK pathways. Research indicates that neutralizing CD44 can specifically inhibit the growth and tumor-initiating capacity of glioma stem-like cells, potentially improving prognosis in patients with high-grade gliomas.
