CD44 shRNA is a gene-silencing tool that utilizes short hairpin RNA (shRNA) to target and reduce the expression of CD44, a cell-surface glycoprotein associated with cancer stem cell (CSC) properties and therapy resistance. In preclinical models of hepatocellular carcinoma (HCC), CD44 shRNA has been shown to disrupt the CD44/mTOR signaling axis, which in turn suppresses autophagy and CSC-mediated survival. This knockdown enhances the sensitivity of HCC cells to radiation therapy (radiosensitivity) and increases radiation-induced apoptosis, suggesting its potential as a therapeutic strategy to overcome radioresistance in advanced or refractory liver cancer.
