Overview

CD44v3 mimetic peptide 4 is a synthetic research-stage peptide designed to disrupt the protein-protein interaction between Tropomyosin receptor kinase A (TrkA) and the variant 3 isoform of the CD44 glycoprotein (CD44v3). In breast cancer models, Nerve Growth Factor (NGF) promotes tumor aggressiveness by inducing the formation of a TrkA/CD44v3 complex. This peptide, which possesses the amino acid sequence IDDDEDFISST (corresponding to the C-terminal region of CD44v3), acts as a decoy that binds to TrkA and prevents its association with CD44v3. Preclinical studies have shown that treatment with this peptide inhibits the proliferation, migration, and invasion of triple-negative breast cancer (TNBC) cells, such as the MDA-MB-231 line, and effectively blocks tumor growth and metastasis in vivo. The peptide was developed and characterized in an academic research setting to explore the therapeutic potential of targeting TrkA/CD44v3 signaling.
