Overview

CD44v6 CAR-T cells are a form of chimeric antigen receptor (CAR) T cell therapy in which patient or donor-derived T lymphocytes are genetically engineered to express a synthetic receptor targeting the variant 6 isoform of the cell surface glycoprotein CD44 (CD44v6). This fourth-generation construct is typically introduced using a lentiviral vector. The therapy is designed to recognize and kill tumor cells expressing the CD44v6 antigen through direct cytotoxicity and immunomodulation. Preclinical studies have demonstrated potent antitumor activity against hematologic malignancies such as acute myeloid leukemia (AML) and multiple myeloma, as well as solid tumors including head and neck squamous cell carcinoma (HNSCC), lung adenocarcinoma, and ovarian carcinoma. The mechanism relies on specific recognition of the tumor-associated antigen CD44v6 while sparing most normal tissues; however, reversible monocytopenia has been observed. Some constructs include safety switches for rapid ablation in case of toxicity[1][3][4][5][2]. Clinical trials are ongoing.
