Overview

CD44v6 monoclonal antibody is an immunotherapeutic agent targeting the CD44v6 splice variant, which is specifically expressed on acute myeloid leukemia (AML) cells but minimally on normal hematopoietic stem cells. CD44v6 promotes AML progression and chemoresistance (to Ara-C and Venetoclax) by driving a transcriptional program through a CD44v6-SPP1 autocrine loop. This signaling axis activates the MAPK/DUSP pathways and shifts anti-apoptotic dependence toward MCL-1 and BCL-XL. Blockade with a CD44v6 monoclonal antibody has been shown to partially reverse these pro-survival effects in preclinical models, making it a promising target for overcoming resistance in high-risk AML.
