CD45 antibody refers to a class of monoclonal antibodies and antibody-based therapeutics that target CD45 (also known as leukocyte common antigen or PTPRC), a receptor-type protein tyrosine phosphatase expressed on all differentiated hematopoietic cells except erythrocytes and platelets. Because of its selective and widespread expression on immune cells and hematopoietic stem cells, CD45 is targeted for various therapeutic and diagnostic applications. In therapeutics, anti-CD45 antibodies (often conjugated to radionuclides like iodine-131 or actinium-225, or to cytotoxic payloads as antibody-drug conjugates) are primarily developed as targeted conditioning agents to deplete host hematopoietic stem cells and immune cells prior to bone marrow transplantation or gene therapy, as well as for treating hematological malignancies like leukemia and lymphoma. In diagnostic and research settings, CD45 antibodies are widely utilized in microfluidic devices (such as the geometrically activated surface interaction chip) for the negative enrichment of circulating tumor cells by depleting white blood cells, as well as in flow cytometry and immunohistochemistry.
