CD45-tethered interleukin-12 is a recombinant fusion protein designed to enhance adoptive T cell therapies (ACT) by localizing cytokine activity to the tumor microenvironment. Developed by Repertoire Immune Medicines (formerly Torque Therapeutics), the biologic consists of the potent immune-activating cytokine interleukin-12 (IL-12) linked to an anti-CD45 antibody fragment (Fab). This construct is designed to be "tethered" to the surface of tumor-specific T cells (such as TILs, CAR-T, or TCR-T cells) via the highly expressed CD45 receptor prior to adoptive transfer. By anchoring IL-12 to the T cell surface, the therapy provides defined cytokine dosages that act in both an autocrine and paracrine manner upon antigen encounter in the tumor, thereby improving antitumor efficacy and repolarizing the immunosuppressive microenvironment while minimizing the severe systemic toxicities typically associated with systemic IL-12 administration.
