Drug intelligence / Profile preview
CD45 Universal Heme ADC (also known as CIM053-SG3376) is a preclinical-stage antibody-drug conjugate (ADC) developed by Cimeio Therapeutics in collaboration with ADC Therapeutics. It targets CD45 (receptor-type tyrosine-protein phosphatase C), a pan-hematopoietic marker highly expressed on both healthy blood cells and hematologic malignancies such as acute myeloid leukemia (AML). The ADC consists of a humanized anti-CD45 monoclonal antibody (CIM053) conjugated via a non-cleavable linker to SG3376, a highly potent pyrrolobenzodiazepine (PBD) dimer payload. To overcome the severe systemic toxicity associated with targeting CD45, the ADC is designed to be administered as a Shielded-Cell & Immunotherapy Pair (SCIP) alongside the transplantation of hematopoietic stem and progenitor cells (HSPCs) that have been gene-edited to express an epitope-modified CD45 variant. This molecular shield protects the healthy transplanted cells from ADC-mediated depletion while allowing the ADC to selectively and completely eradicate diseased CD45-positive leukemic cells.
