Overview

This investigational cellular therapy, developed by St. Jude Children's Research Hospital, is a specialized form of haploidentical (half-matched) hematopoietic progenitor cell transplantation (HPCT) designed for pediatric patients with high-risk or relapsed/refractory hematologic malignancies. The approach involves the selective depletion of CD45RA+ naive T cells from the donor graft to minimize the risk of graft-versus-host disease (GvHD) while preserving CD45RO+ memory T cells to facilitate rapid immune reconstitution and protection against viral infections. This is combined with the infusion of donor-derived natural killer (NK) cells to enhance the graft-versus-leukemia (GvL) effect. The therapy is typically administered following a reduced-intensity conditioning regimen and is being evaluated for conditions such as acute lymphoblastic leukemia (ALL), acute myeloid leukemia (AML), and myelodysplastic syndrome (MDS).
