CD47 CAR macrophage is an engineered cell therapy designed to target CD47, a widely expressed immune checkpoint often referred to as a "don't eat me" signal on tumor cells. This specific iteration, developed by researchers at Xidian University and collaborating institutions, utilizes a tumor microenvironment (TME)-responsive platform. It incorporates a CD47-specific single-chain variable fragment (scFv) and a costimulatory domain derived from the α1β1 integrin-mediated Fc-gamma receptor I (FcγRI) signaling component. To mitigate systemic toxicity—particularly against erythrocytes which also express CD47—the expression of the CAR is controlled by an arginase 1 (Arg1) promoter, ensuring activation primarily within the immunosuppressive TME. This "Trojan horse" approach aims to enhance phagocytosis and antitumor activity against solid tumors while minimizing off-target effects.
