The CD47 IgG2σ antibody is a research-stage monoclonal antibody targeting CD47 (integrin-associated protein), a 'don't eat me' signal overexpressed on tumor cells. It is engineered with a human IgG2σ (sigma) Fc domain, which is effector-silent, meaning it does not bind to Fcγ receptors. This design aims to block the CD47–SIRPα interaction to promote macrophage-mediated phagocytosis (ADCP) when combined with other opsonizing antibodies, while avoiding the hemagglutination and red blood cell depletion often associated with wild-type Fc CD47 antibodies. It has been investigated primarily in hematologic malignancies such as multiple myeloma and leukemia.
