CD47 scFv is a single-chain variable fragment (scFv) designed to bind to and block CD47, a cell surface protein that functions as a "do not eat me" signal overexpressed on various malignant tumors, including multiple myeloma. By blocking the interaction between CD47 on tumor cells and SIRP-α on macrophages, it aims to promote tumor phagocytosis by the innate immune system. In research conducted at the Medical College of Wisconsin, this scFv has been utilized both as a standalone agent and as a modular component for generating bispecific antibodies, such as a CD47-CD138 construct. This approach is intended to improve tumor selectivity and reduce off-target toxicities associated with the widespread expression of CD47 on normal tissues, thereby mitigating the "sink effect" and enhancing therapeutic efficacy.
