**CD4CAR NK-92** is a preclinical, third-generation anti-CD4 chimeric antigen receptor natural killer cell therapy. It consists of clonal NK-92 cells genetically engineered with a CD4-targeting CAR containing an ibalizumab-derived anti-CD4 single-chain variable fragment and CD28, 4-1BB, and CD3ζ signaling domains. The therapy is designed to selectively eliminate CD4-expressing malignant cells and has shown ex vivo cytotoxicity against CD4-positive peripheral T-cell lymphoma, T-cell acute lymphoblastic leukemia, Sézary syndrome, and other CD4-expressing leukemia and lymphoma models, as well as antitumor activity in a KARPAS-299 xenograft model. Development has been associated with iCell Gene Therapeutics and Stony Brook University research groups; no marketed product, INN, or registered clinical development program for this exact NK-92 product was identified.
