CD5 CAR-NK is a genetically engineered cell therapy product consisting of natural killer (NK) cells modified to express a chimeric antigen receptor (CAR) targeting CD5, a cell surface glycoprotein highly expressed on malignant T-cells and some B-cell lymphomas. Unlike T cells, NK cells do not express CD5, making them an attractive platform to minimize self-targeting (fratricide) in CAR therapy for T-cell malignancies. These engineered NK cells exhibit potent, targeted cytotoxicity against CD5-positive malignancies, including T-cell acute lymphoblastic leukemia (T-ALL), peripheral T-cell lymphomas (PTCL), Sézary syndrome, chronic lymphocytic leukemia (CLL), and some B-cell lymphomas. Development platforms include both primary peripheral blood-derived NK cells and immortalized NK-92 cell lines. Some iterations integrate suicide gene systems (such as HSV-TK) for enhanced safety. Preclinical and early-phase clinical data demonstrate robust anti-tumor efficacy and an acceptable safety profile, providing a promising option for relapsed or refractory T-cell hematological malignancies[1][2][4].
