Overview

ICT-CD5 (CD5 CAR T) is an autologous chimeric antigen receptor (CAR) T-cell therapy developed by iCell Gene Therapeutics for the treatment of aggressive T-cell malignancies. The therapy targets CD5, a surface glycoprotein constitutively expressed on normal T cells and most T-cell leukemias and lymphomas, such as T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoblastic lymphoma (T-LBL). A primary challenge in targeting CD5 is "fratricide," where the CAR T cells attack each other because they also express the CD5 antigen. iCell utilizes a proprietary non-gene-editing platform to overcome this issue, likely employing protein expression blockers (PEBL) to sequester or downregulate CD5 protein expression on the CAR T cell surface without the need for genomic disruption via CRISPR or other editing tools. This approach preserves the integrity of the T-cell genome while enabling the cells to persist and target malignant T cells effectively.
