Overview

CD5 CAR T-cell is an investigational autologous or allogeneic chimeric antigen receptor (CAR) T-cell therapy developed by iCell ImmunityX (iCell Gene Therapeutics) for the treatment of T-cell malignancies, including T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoma. The therapy targets CD5, a 67-kDa surface glycoprotein expressed on the majority of mature T cells and T-cell neoplasms. A significant challenge in developing CAR T cells for T-cell malignancies is "fratricide," where the CAR T cells target and kill each other because they naturally express the target antigen. iCell utilizes a proprietary non-gene-editing technology, such as a protein expression blocker (PEBL), to downregulate or sequester surface CD5 expression on the CAR T cells themselves. This approach prevents fratricide and allows for the expansion and persistence of the therapeutic cells without the need for genomic disruption via CRISPR or other gene-editing tools. The drug is currently in Phase 1 clinical development.
