Overview

CD5 KO anti-mesothelin CAR T cells are an investigational adoptive cell therapy developed by researchers at the University of Pennsylvania. This therapy utilizes chimeric antigen receptor (CAR) T cells engineered to target mesothelin (MSLN), a cell-surface glycoprotein overexpressed in various solid tumors, including pancreatic ductal adenocarcinoma (PDAC), mesothelioma, and ovarian cancer. To enhance the potency and persistence of the T cells within the immunosuppressive tumor microenvironment, the CD5 gene—a transmembrane protein that acts as a negative regulator of T cell receptor signaling—is knocked out using CRISPR/Cas9 technology. Preclinical studies indicate that this 'disinhibition' strategy leads to significantly increased T cell expansion, improved tumor control, and prolonged overall survival compared to standard mesothelin-targeted CAR T cells.
