Overview

CD56 CAR T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target Neural Cell Adhesion Molecule 1 (NCAM-1), also known as CD56. CD56 is a cell surface glycoprotein highly expressed in malignancies with neuronal or neuroendocrine differentiation, including multiple myeloma, neuroblastoma, small-cell lung cancer (SCLC), and certain types of leukemia and lymphoma. The therapy involves the genetic modification of T cells (either autologous or allogeneic) to express a CAR consisting of an anti-CD56 single-chain variable fragment (scFv) fused to intracellular signaling domains, typically including CD3ζ and a costimulatory domain such as 4-1BB or CD28. This engineering enables the T cells to recognize and kill CD56-positive tumor cells. Notably, CD56 CAR T cells have shown the ability to avoid significant fratricide despite the expression of CD56 on some activated T cells. Research is primarily conducted by academic institutions such as Memorial Sloan Kettering Cancer Center and the Shenzhen Geno-Immune Medical Institute.
